In vitro culturing of plant cells can cause changes in karyotype. Chromosome variations following long-term propagation in suspension culture of Triticum timopheevii (Zhuk.) Zhuk. were studied by routine staining and C-banding. The culture was highly heterogeneous with respect to the number and structure of chromosomes. The modal class cells had a lower chromosome number than T. timopheevii (2n= 28). This data was confirmed by cytophotometric analysis of nuclear-DNA content. Frequencies of chromosome loss varied for different homoeologous groups. At genome chromosomes tended to be preferentially eliminated in cells of different ploidy levels. Deletions, insertions, translocations, telocentric chromosomes, isochromosomes and dicentrics and their derivatives were observed in cultured cells. Chromosomes of various homoeologous groups differed in the frequencies and spectra of re—arrangements, but most aberrations occurred in the G-genome chromosomes. In vitro chromosome modifications did not correspond to in vivo variation. Presumably, this difference was caused by differences in the mechanisms of adaptation to the environment at the levels of the cell and the whole organism. G-genome chromosomes were more frequently involved in this process, both in vivo and in vitro. 相似文献
A method for PEG treatment of bovine sera to be used for cell cultures is presented. The procedure consists in repeated freezing and thawing cycles of the sera, after PEG precipitation until a clear solution is obtained. The sera, so prepared, are homogeneous in composition and appropriate for cell growth in primary cell cultures and cell lines. 相似文献
Recent research has shown that cattle that develop mucosal disease (MD) often, if not always, have been persistently infected with bovine viral diarrhea virus (BVDV) since birth. The purpose of the present study was to determine whether MD could be induced by immunosuppression of persistently BVDV-infected cattle. For that purpose, adrenocorticotropic hormone (ACTH) was injected intramuscularly, twice daily for 5 consecutive days in 4 persistently BVDV-infected cattle and in 3 control cattle. Before the ACTH treatment, the numbers of leukocytes, neutrophils and mononuclear cells (MNC) per litre of blood in BVDV-infected cattle were in the same range as in the controls. Similarly, the proportions of B cells, T cells, monocytes and Fcγ+ cells (cells with receptor for the Fc part of IgG) were the same in the 2 groups of animals. On the other hand, the proliferative response to mitogen stimulation of MNC obtained from the control animals was twice as high as the corresponding value of the persistently BVDV-infected cattle.In all animals, ACTH treatment caused increased Cortisol concentrations, leukocytosis, neutrophilia and decreased mitogen-induced lymphocyte stimulation. However, the MNC count and the proportions of B cells, T cells, Fcγ+ cells and monocytes remained unaltered. In spite of the immunosuppression, indicated by the decrease in mitogen-induced lymphocyte stimulation. Conditioned medium prepared from the mouse fibroblast cell line L 929 which is known to produce colony-stimulating factor active on mouse bone marrow cells was also able to stimulate the growth of swine bone marrow cells in a liquid culture system. During the first 4 days of culture mononuclear phagocyte and granulocyte colonies developed. Prolonged cultured cells were classified belonging to the macrophage lineage by morphological and cytochemical criteria. These cells fulfill also functional characteristics for macrophages, like expression of Fc receptors, immune phagocytosis and production of prostaglandins. A sheep-pox virus strain has been adapted and multiplied in primary lamb kidney cell cultures. The main characteristics of the strain have been verified in vitro after clones were isolated, and the results confirmed its identity. The safety and the potency of the strain have been investigated in sheep.
The inoculation of the strain to sheep was followed by a post-vaccinal reaction materialised by a nodule at the site of inoculation and an increase of temperature by about 1°C. No reactions adversely affecting pregnancy have been noted. Immunisation was demonstrated by an increase in the level of neutralising serous antibodies and protection against the pathogenic virus. The immunity tended to decrease during the second year after primovaccination and a yearly booster vaccination appeared to be necessary. Primovaccination of lambs over 2 months of age produced a better immunity, especially when the lambs were born from vaccinated ewes.
This strain forms the active principle of a freeze-dried vaccine containing no adjuvant of the immunity. 相似文献
ansitional cell urethral carcinoma in four female dogs is reported. Lesion distribution in the proximal two thirds of the urethra and the radiographic appearance with retrograde positive contrast urethrography were similar in each dog. The normally smooth longitudinal urethral striations were replaced by multiple small, poorly marginated intraluminal masses that resulted in a generalized moth-eaten appearance. Extension of contrast medium into the periurethral tissues also occurred. Retrograde urethrography, a simple diagnostic technic, aided in establishing a differential diagnosis in dogs that had urine retention and stranguria. 相似文献

Abstract— Epidermotropic cutaneous T-cell lymphoma was diagnosed in six Syrian hamsters (Mesocricetus auratus) with a generalised dermatosis characterised by pruritus, alopecia and exfoliative erythroderma. Plaques and nodules were evident in one animal. Clinical and histopathological findings were suggestive of mycosis fungoides and electron microscopical investigations in one case were supportive. Immuno-histochemical stains for CD3 antigen were strongly positive in all cases. Résumé— Un lyphome T cutané a été diagnostiqué chez six hamsters de Sibérie (Mesocricetus auratus) avec une dermatose généralisée, caractérisée par un prurit, une alopécle et une érythrodermie exfoliative. Des plaques et des nodules ont été observées sur un animal. Les données cliniques et histopathologiques étaient évocatrices d'un mycosis fungoïdes et les examens en microscopie électronique étaient compatibles dans un cas. Les marquages immuno-histochimiques de l'antigène CD3 étaient très positifs dans tous les cas. Los estudios clinicos e histopatológicos indicaron una micosis fungoide, y las investigaciones de microscopía, en uno de los caseos, pareció indicar lo mismo. 15N-aided investigations were conducted to ascertain the Nj fixation and the nitrogen (N) contribution by mungbean ( Vigna radiata L.) and groundnut ( Aracbis hypogaea L.) when intercropped with maize ( Zea mays ). The study involved growing seven genotypes of the above legumes with maize in alternate rows in two separate experiments. A sole crop of maize was used as the reference crop to determine N2 fixation by the 15N methodology. Significant genotypic differences in pod yield and stover N content were observed in intercropped mungbean and groundnut. The percentage N derived from the atmosphere showed a genotypic variation of 31 to 45 % (7 to 10 kg N2 fixed ha−1O in mungbean and 47 to 69 % (9 to 18 kg N2 fixed ha−1) in groundnut. Harvest index for N varied from 58 to 77 % in mungbean and 55 to 75 % in groundnut. In groundnut, the uptake of soil N was significantly affected by the genotype. Assuming that the N contribution to the soil by the helow-ground plant parts was negligible, the removal of seeds at maturity resulted in a negative N balance in the soil in all the genotypes of mungbean. In groundnut, some genotypes produced a positive N balance in the soil. Owing to high N2 fixation capacity and low harvest index for N, groundnut showed a greater N supplementing ability than mungbean. 相似文献
